The preparation methods utilized by manufacturers worldwide of NAD powder are mainly divided into chemical synthesis method and biocatalytic method, among which biocatalytic method includes biological fermentation method and enzyme catalysis method. Enzyme catalysis method has gradually become the mainstream direction because of its advantages of green, environmental protection and pollution-free. And then the purity of NAD powder will reach 99% after the procedure of further purifying. But some NAD manufacturers tend to choose the multiple methods to develop the producing of NAD.
Molecules that can be taken in supplement form to increase NAD levels in the body are referred to by some as “NAD boosters.” Studies conducted over the past six decades suggest that the following are some of the many benefits associated with taking an NAD supplement:
Can Help Restore Mitochondrial Function
Helps Repair Blood Vessels —A 2018 mice study found that supplementation could aid in repair and growth of aged blood vessels. There’s also some evidence it can help manage heart disease risk factors like high blood pressure and high cholesterol.
May Improve Muscle Function — One animal study conducted in 2016 found that degenerative muscles had improved muscle function when supplemented with NAD+ precursors.
Potentially Helps Repair Cells and Damaged DNA — Some studies have found evidence that NAD+ precursor supplementation leads to an increase in DNA damage repair. NAD+ is broken down into two component parts, nicotinamide and ADP-ribose, which combine with proteins to repair cells.
May Help Improve Cognitive Function — Several studies conducted on mice have found that mice treated with NAD+ precursors experienced improvements in cognitive function, learning and memory. Findings have led researchers to believe that NAD supplement may help protect against cognitive decline/Alzheimer’s disease.
May Help Prevent Age-Related Weight Gain — A 2012 study showed that when mice fed a high-fat diet were given an NAD supplement, they gained 60 percent less weight than they did on the same diets without the supplement. One reason this may be true is that nicotinamide adenine dinucleotide helps regulate production of stress- and appetite-related hormones, thanks to its effects on circadian rhythms.
Precursors are molecules used in chemical reactions inside the body to create other compounds. There are a number of precursors of NAD+ that result in higher levels when you consume enough of them.
1.Prevention and treatment of viral-induced inflammatory storms
Scientists have found after extensive research that neo-coronavirus has a mechanism similar to SARS virus to activate inflammatory vesicles NLRP3. and the activation of NLRP3 produces more inflammatory factors, generating excessive inflammation and thus triggering a deadly cytokine storm. This problem can be well addressed by NAD+, which inhibits the activity of NF-κB inflammatory pathway and NLRP3 inflammasome by increasing the activity of sirtuins (SIRT1, SIRT2 and SIRT3), thus preventing cytokine storm caused by excessive inflammation. Therefore, Sinclair and other scientists believe that increasing the concentration of NAD+ may play an important role in the prevention and treatment of neocoronavirus and other viral infections.
2.Restoration of virus-induced metabolic disorders
NAD+ is an essential coenzyme for many cellular energy metabolic pathways, present in every cell of the body, involved in thousands of reactions, and an important player in maintaining cellular viability. In the COVID-19 infection model, NAD+ and NMN supplementation was found to be effective in alleviating cell death and protecting the lung.
First, inspect the factory. After some screening, NAD manufactures that directly face consumers pay more attention to brand building. Therefore, for a good brand, quality is the most important thing, and the first thing to control the quality of raw materials is to inspect the factory. Bontac company actually manufacturing NAD powder of high quality with the caterias of SGS. Secondly, the purity is tested. Purity is one of the most important parameters of NAD powder. If high purity NAD cannot be guaranteed, the remaining substances are likely to exceed the relevant standards. As the attached certificates demonstrates that the NAD powder produced by Bontac reach the purity of 99.9%. Finally, a professional test spectrum is needed to prove it. Common methods for determining the structure of an organic compound include Nuclear Magnetic Resonance Spectroscopy (NMR) and high-resolution mass spectrometry (HRMS). Usually through the analysis of these two spectra, the structure of the compound can be preliminarily determined.
In the international market, NAD+ food supplements are in production in the United States, the Australian Food and Drug Administration has approved NAD+ products for sale, and Japanese NAD+ manufactures are devoted to extend the market share in China. Li Ka-shing invested HK$200 million in the US NAD manufacturer, whose NAD+ products are being sold by Watsons in Hong Kong! Then Mclane, a company owned by Warren Buffett also joined the NAD market.
1. Introduction The gut microbiota has long been regarded as one of the key elements contributing to the regulation of host health. Any changes in the composition or quality of the gut microbiota may have physiological consequences for the host. To determine the effect of sweetener stevia (also known as stevioside) on the gut microbiome of healthy population, the stool samples are collected from healthy participants who consume with or without five drops of the sweetener stevia twice daily. Following analyses of 16S rRNA sequencing method, no large-scale change is found in the gut microbiota post 12 weeks of consumption with stevia, hinting the safety of stevia. 2. Insignificant changes in the alpha or beta diversity following consumption of stevia It is discovered that there is no significant difference in alpha diversity (in terms of observed taxa, evenness and Shannon Index) and beta diversity (with regard to PCoA, PERMANOVA, and Jaccard Index) between groups. Nevertheless, PCoA plots shows strong separation along the x-axis. In addition, the community composition in each group is relatively even over time and equally diverse. 3. No clear difference in relative abundances of taxa At the genus level, relative abundances are similar between the control and stevia groups. No major difference is observed in relative abundances at the class, order and family level. Strikingly, butyricoccus is the only one identified taxon exhibiting significant difference at baseline, but not after 12 weeks of stevia consumption. Moreover, Collinsella and Aldercreutzia are two coprococcus species identified as explicitly different at baseline (one higher and one lower when comparing stevia vs. control), which however are significantly elevated after 12 weeks of consumption with stevia. 4. The safe intake volume of sweetener steviol glycosides In the European Food Safety Authority (EFSA), there is a Panel on Food Additives and Flavourings (FAF), which is responsible for evaluating the safety of food additives and establishing acceptable daily intake levels for safe use. Steviol glycosides, one of the extract from stevia, is evaluated by the FAF as well. In accordance to the latest toxicological test, this sweeter is not genotoxic and carcinogenic, without any adverse effects on the human reproductive system or growing children. The expert group has set the acceptable daily intake (ADI) of steviol glycosides at 4 milligrams per kilogram of body weight per day, which is consistent with the level determined by the Joint Expert Committee on Food Additives (JECFA) administered by the US Food and Agriculture Organization (FAO) and the World Health Organization (WHO). 5. Conclusion Regular, long-term consumption of stevia does not overtly alter the composition of the human gut microbiotia. Stevia can be safe as long as the intake volume is controlled appropriately. 1. Introduction Age-related NAD+ depletion affects physiological functions and contributes to various aging-related diseases. NAD+ precursors can significantly elevate NAD+ level in murine tissues, effectively mitigate metabolic syndrome, enhance cardiovascular health, protect against neurodegeneration, and boost muscular strength, with broad prospect in the anti-aging-related field. 2. The synthesis and metabolism of NAD+ in age-related pathologies NAD+ is synthesized from NAD+ precursors and amino acids tryptophan via three main pathways: De novo, Preiss-Handler, and Salvage. Supplementation of NAD+ precursors can be advantageous in maintaining normal cellular metabolism regulated by NAD+ and NAD+-dependent enzymes such as Sirtuins, PARP, CD38, and SARM1. NAD+ intermediates require conversion into NA to elevate NAD+ level. NAD+ and its metabolism-related enzymes have very important roles in biological processes such as cellular metabolic processes, gene expression, apoptosis and carcinogenesis. NAD+ repletion is drawing attention as an anti-aging intervention. NAD+ precursors, such as NA, NAM, NR, and NMN, provide beneficial effects in various preclinical disease models of age-induced deficits, including metabolic disorders, cardiovascular, neurodegenerative diseases, and musculoskeletal diseases. 3. Comparison on the efficacy of replenishing NAD precursors in pre-clinical studies and clinical studies in age-related pathologies The downregulation of NAD+ level in cells and tissues is not a universal phenomenon for aging-related pathologies. NAD+ merely decreases with age in certain tissues. The efficacy of NAD+ precursors in clinical studies has been limited in comparison with that in the pre-clinical studies. Noteworthily, this issue can be addressed as long as much attention has been paid to the metabolism of NAD. With regards to the oral supplementation of NAD+ precursors, there is obvious link between NAD metabolism and gut microbes. Specifically, oral consumption of NMN is converted into NAMN through interaction with the gut microbiome. In addition, dietary NAM and NR are converted into NA through gut microbiota. 4. Future research directions regarding the NAD+ metabolism It is fundamental to consider how the gut microbiome affects NAD+ metabolism, and changes in microbiome composition may affect the availability of NAD+ precursors. Future studies also require the comparative analysis of different precursors, and the role of gut microbiomes regarding various intermediaries needs to be investigated. Assessment of how NAD+ precursors affect microbiota and how their interaction with NAD+ metabolism benefits the physiological condition is essential for future preclinical and clinical studies. 5. Conclusion Supplementation of suitable NAD+ precursors or intervening in NAD+ metabolism can restore the body's NAD+ level, which is of great practical significance for effectively improving aging-related diseases and prolonging healthy life span is of great practical significance for effectively improving aging-related diseases and prolonging healthy life span. NAD metabolism involves gut microbiome, and in-depth research on their interaction is possibly an important breakthrough in the future to combat aging-related pathologies. 1. Introduction Diabetic peripheral neuropathy (DPN) is one of the most frequent complications of diabetes, ans also a major cause of foot ulcers, disability, and eventual amputation. With the prolongation of the diabetes, about 50% of people with diabetes will eventually develop DPN. Notably, supplementing NAD+ precursors could alleviate DPN symptoms by increasing the NAD+ level and activating the sirtuin-1 (SIRT1) protein. 2. The reversal effect of NAD+ precursors on DPN In vitro, the Dorsal Root Ganglion neurons (DRGs) isolated from diabetic mice are exposed to the NAD+ precursor Nicotinamide Riboside (NR) or Nicotinamide Mononucleotide (NMN). It is found that the NAD+ level, the SIRT1 protein, and the deacetylation activity are elevated, followed by the boosted neurite growth, the improved nerve function, and the reversal of IENFD loss. In vivo, supplement of NMN or NR also offsets the neuropathy in C57BL6 mice induced by streptozotocin (STZ) or high fat diet (HFD), as manifested by the improved sensory function, normalized nerve conduction velocities, and restored intraepidermal nerve fibers. 3. The increase of neurite length in a SIRT1-dependent manner post the addition of NMN/NR SIRT1, one of the most unique NAD+ consuming enzymes, can protect against DPN when activated, which may attribute to the improved mitochondrial function and energy homeostasis. Apart from these, SIRT1 activity in the nucleus can deacetylate the transcriptional and co-transcriptional factors that regulate glucose homeostasis and fat oxidation. The activation of SIRT1 is critical for axonal regeneration. NMN/NR treatment or transfection with SIRT1 overexpression vector can directly facilitate the neurite growth in cultured DRG neurons, which however is hindered by the SIRT1 inhibitor EX527, hinting the significance of SIRT1. 4. The association of SARM1 with NMNAT2 in axonal degeneration of DPN Sterile alpha and Toll/interleukin-1 receptor motif-containing 1 (SARM1) controls the axonal degeneration and regeneration via a well-regulated system comprising NAD+ and NMN. NAD and NMNAT2 can boost vesicular glycolysis and axonal transport to maintain the axonal health. The mitochondrial localization of SARM1 complements the coordinated activity of NMNAT2 that promotes axonal survival. 5. Conclusion Supplementing NAD+ precursors may be a promising approach for the treatment of DPN. A SARM1 inhibitor coupled with either NR or NMN may be more effective than a single agent alone in preventing or treating DPN. Reference Chandrasekaran K, Najimi N, Sagi AR, et al.
