Molecules that can be taken in supplement form to increase NAD levels in the body are referred to by some as “NAD boosters.” Studies conducted over the past six decades suggest that the following are some of the many benefits associated with taking an NAD supplement:
Can Help Restore Mitochondrial Function
Helps Repair Blood Vessels —A 2018 mice study found that supplementation could aid in repair and growth of aged blood vessels. There’s also some evidence it can help manage heart disease risk factors like high blood pressure and high cholesterol.
May Improve Muscle Function — One animal study conducted in 2016 found that degenerative muscles had improved muscle function when supplemented with NAD+ precursors.
Potentially Helps Repair Cells and Damaged DNA — Some studies have found evidence that NAD+ precursor supplementation leads to an increase in DNA damage repair. NAD+ is broken down into two component parts, nicotinamide and ADP-ribose, which combine with proteins to repair cells.
May Help Improve Cognitive Function — Several studies conducted on mice have found that mice treated with NAD+ precursors experienced improvements in cognitive function, learning and memory. Findings have led researchers to believe that NAD supplement may help protect against cognitive decline/Alzheimer’s disease.
May Help Prevent Age-Related Weight Gain — A 2012 study showed that when mice fed a high-fat diet were given an NAD supplement, they gained 60 percent less weight than they did on the same diets without the supplement. One reason this may be true is that nicotinamide adenine dinucleotide helps regulate production of stress- and appetite-related hormones, thanks to its effects on circadian rhythms.
Precursors are molecules used in chemical reactions inside the body to create other compounds. There are a number of precursors of NAD+ that result in higher levels when you consume enough of them.
1.Prevention and treatment of viral-induced inflammatory storms
Scientists have found after extensive research that neo-coronavirus has a mechanism similar to SARS virus to activate inflammatory vesicles NLRP3. and the activation of NLRP3 produces more inflammatory factors, generating excessive inflammation and thus triggering a deadly cytokine storm. This problem can be well addressed by NAD+, which inhibits the activity of NF-κB inflammatory pathway and NLRP3 inflammasome by increasing the activity of sirtuins (SIRT1, SIRT2 and SIRT3), thus preventing cytokine storm caused by excessive inflammation. Therefore, Sinclair and other scientists believe that increasing the concentration of NAD+ may play an important role in the prevention and treatment of neocoronavirus and other viral infections.
2.Restoration of virus-induced metabolic disorders
NAD+ is an essential coenzyme for many cellular energy metabolic pathways, present in every cell of the body, involved in thousands of reactions, and an important player in maintaining cellular viability. In the COVID-19 infection model, NAD+ and NMN supplementation was found to be effective in alleviating cell death and protecting the lung.
First, inspect the factory. After some screening, NAD manufactures that directly face consumers pay more attention to brand building. Therefore, for a good brand, quality is the most important thing, and the first thing to control the quality of raw materials is to inspect the factory. Bontac company actually manufacturing NAD powder of high quality with the caterias of SGS. Secondly, the purity is tested. Purity is one of the most important parameters of NAD powder. If high purity NAD cannot be guaranteed, the remaining substances are likely to exceed the relevant standards. As the attached certificates demonstrates that the NAD powder produced by Bontac reach the purity of 99.9%. Finally, a professional test spectrum is needed to prove it. Common methods for determining the structure of an organic compound include Nuclear Magnetic Resonance Spectroscopy (NMR) and high-resolution mass spectrometry (HRMS). Usually through the analysis of these two spectra, the structure of the compound can be preliminarily determined.
In the international market, NAD+ food supplements are in production in the United States, the Australian Food and Drug Administration has approved NAD+ products for sale, and Japanese NAD+ manufactures are devoted to extend the market share in China. Li Ka-shing invested HK$200 million in the US NAD manufacturer, whose NAD+ products are being sold by Watsons in Hong Kong! Then Mclane, a company owned by Warren Buffett also joined the NAD market.
1.Introduction Mitochondria, a type of highly plastic organelle, can maintain a relatively stable dynamic equilibrium state towards the morphology, structure, quantity, volume, quality, and function under normal physiological conditions. This dynamic equilibrium state is generally called mitochondria homeostasis. Replenishment of nicotinamide mononucleotide (NMN) to boost intracellular NAD+ level can regulate mitochondrial homeostasis. 2.The amelioration of mitochondrial dysfunction and senescence post NMN treatment Typically, NAD+ level determines the rate and extent of senescence. The NAD+ pool experienced by cells and tissues presents a declined tendency with age, which may lead to a loss of mitochondrial homeostasis and senescent phenotypes in CD8+ T cells. Following NMN (100 μM) supplementation, NAD+ level is restored and the NAD+/NADH ratio is elevated in senescent CD8+ T cells, by which the mitochondrial functions are obviously improved and the cytoplasmic mtDNA is reduced. 3.The prevention of neuroinflammation and senescence in CD8+ T cells via NMN supplement Post NMN supplementation, the senescence and neuroinflammation are correspondingly repressed. With a great detail, NMN supplementation decreases the ROS and mitochondrial membrane potential in CD8+ T cells, while increasing the intracellular ATP. Furthermore, NMN suppresses the inflammation-associated markers (IL-1β, IL-6, TNF-α and IFN-γ) in the supernatant and the mRNA of senescent CD8+ T cells. 4. The inhibitory effect of NMN on the senescence-related pathway STING The activation of STING can launch a robust proinflammatory response and senescence, which is closely related to the deficient health span in model mice. Strikingly, NMN can prominently improve the senescent state of T cells and reduce the secretion of senescence-related inflammatory factors via inhibition of the STING pathway. 5. Conclusion Upregulation of NAD+ level by NMN supplementation can regulate the mitochondrial homeostasis to prevent STING-induced senescence in CD8+ T cells. Reference Ye B, Pei Y, Wang L, et al. 1.Introduction Nicotinamide adenine dinucleotide phosphate hydrogen (NADPH), also known as reduced coenzyme II, is a critical cofactor in cellular antioxidant systems and lipid synthesis, which links insulin resistance and ferroptosis of pancreatic β cells in the context of metabolic disorders such as diabetes mellitus, playing a central role in maintaining metabolic homeostasis. 2. Biological role of NADPH NADPH functions as a coenzyme essential to cellular metabolism, playing pivotal roles in various critical biological processes, such as ROS scavenging, ROS production, fatty acid synthesis and cholesterol synthesis. 3. Biosynthetic pathway of NADPH Cellular production of NADPH is facilitated through several pathways, including the pentose phosphate pathway, the citric acid cycle, and fatty acid metabolism. The dynamic equilibrium between NADPH synthesis and consumption is essential for preserving cellular redox balance and enabling a host of biosynthetic reactions. 4. The role of NADPH in insulin secretion from pancreatic β-Cells Both redox reaction and metabolic signaling can modulate insulin secretion from pancreatic β-cells, where NADPH plays a central role. It can not only serves as a metabolic coupling factor, but also acts as a custodian of β-cell integrity, delicately managing the interplay between metabolic inputs and insulin output. 5. The interaction between insulin resistance and NADPH A substantial body of evidence reveals that NADPH is critical for the regulation of oxidative stress and inflammatory responses, the main contributors to the pathogenesis of insulin resistance. Specifically, NADPH is implicated in ROS production via NOX and is also utilized in the synthesis of new fatty acids, which contributes to the development of insulin resistance, particularly in the context of obesity-induced chronic inflammation. 6. The impact of NADPH upon the ferroptosis in the context of diabetes In pancreatic β cells, the elevated blood sugar and pro-inflammatory cytokines can trigger oxidative stress and iron accumulation to promote lipid peroxidation, thereby facilitating the ferroptosis. In return, the ferroptosis can reduce insulin secretion and beta cell mass, which is contributive to the progression of diabetes. In general, NADPH plays a dual role in ferroptosis. On the one hand, it can promote ROS generation via NOX. On the other hand, it can support antioxidant defense through glutathione regeneration. In the context of diabetes, NADPH may predominantly fuel processes leading to ferroptosis, mainly due to the enhanced activity and affinity of NOX, which however requires further research for verification. 7. Conclusion NADPH has a critical role in the complex landscape of metabolic disorders, particularly insulin resistance and ferroptosis. Regulating NADPH-related pathways may open up new opportunities for the treatment of metabolic disorders. Introduction Triocresyl phosphate (TOCP) is widely used in the realm of industry and agriculture in the last century. However, it is subsequently banned due to the increasing understanding of its toxicity. In the 21st century, TOCP comes back into the limelight as the aviation industry springs up. This research uncovers the adverse effects of TOCP on the reproductive system. Notably, nicotinamide mononucleotide (NMN), a crucial intermediate in the generation of NAD+, may serve as a therapeutic intervention to attenuate the oocyte damage caused by TOCP. About TOCP TOCP, a classic aromatic organophosphate ester, generally functions as flame retardant, plasticizer, lubricant, and jet fuel additive due to its chemical and thermal stability. At room temperature, TOCP is an odorless, yellowish transparent liquid. It is insoluble in water, but soluble in organic solvents such as alcohol, ether and benzene. In addition to its use in aviation industry, TOCP is currently applied in the manufacturing of construction materials such as plastics, furniture, textiles, printed circuit boards, and insulation. The negative roles of TOCP in oocytes Through the analyses of germinal vesicle breakdown (GVBD) and polar body extrusion (PBE), it is discovered that TOCP impedes the maturation process of oocyte meiotic division, suppressing the reinitiation of oocytes and the final extrusion of the first polar body. Remarkably, maturation of oocytes is deemed as a critical prerequisite for successful fertilization and subsequent embryonic development. Besides, it triggers disturbances in the cytoskeleton of oocytes and affects the distribution and functionality of mitochondria. Furthermore, exposure to TOCP alters the genes related to histone modification in oocytes, as manifested by the elevated levels of histone methylation at H3K9me3 and H3K27me3. The reversing effects of NMN on TOCP in oocytes Replenishing NMN partially restores the spindle/chromosome structure as well as the attachment of microtubules to centromeres, and stabilizes the distribution of actin filaments, thereby maintaining chromosomal integrity and supporting the nuclear maturation process of oocytes. Meanwhile, NMN is also effective in rescuing mitochondrial dysfunction induced by TOCP, which restores membrane potential and ATP levels, reduces excessive ROS production, prevents DNA damage, and hinders cell apoptosis as well as epigenetic alterations. Conclusion Nicotinamide mononucleotide maintains cytoskeletal stability and fortifies mitochondrial function to mitigate oocyte damage induced by TOCP, signifying its potential application value in refining reproductive therapeutic strategies.
