1. Introduction Nicotinamide mononucleotide (NMN) supplementation has been suggested to hamper the inflammatory responses via restoring NAD+ level and downregulating the expression of Cyclooxygenase-2 (COX-2). Strikingly, both Aryl hydrocarbon receptor (AhR) and Indoleamine 2,3-Dioxygenase 1 (IDO1), two key enzymes for kynurenine production, can mediate the anti-inflammatory function of NMN in RAW 264.7 macrophages. 2. The alleviated inflammatory response in the presence of NMN supplementation For deciphering the impact of NMN in vivo, mice are subjected to daily intraperitoneal (i.p.) injection of NMN (500 mg/kg) for consecutive 6 days, followed by i.p. injection of lipopolysaccharides (LPS) (5 mg/kg) or alum (700 μg) on day 7. A Scientific Method for Quantifying NMN in Biological Samples

1. Introduction Supplementation of nicotinamide mononucleotide (NMN) to upregulate the level of nicotinamide adenine dinucleotide (NAD+) has been unveiled to be a promising anti-aging intervention. However, it is still a serious challenge to accurately quantify NAD+ intermediates, NMN in particular. This study is powered to introduce a novel method, double isotope-mediated LC-MS/MS methodology (dimeLC-MS/MS), for the precise quantification of NMN in biological samples. 2. Factors affecting the accurate detection of NMN NMN is hard to be accurately detected due to its vulnerability to enzymatic degradation, conversion in sample processing, its complex behaviors in different column and extraction conditions, as well as matrix effect. Specifically, NMN has the properties of high polarity and low volatility, which is easy to dissolve in water but difficult to dissolve in organic solvents. These properties greatly restrict the application of many conventional quantitative analysis methods. Biological samples such as blood carries significant activities of CD38 and CD73 (ecto-5’-nucleotidase), both of which could use NMN as a substrate. The behavior of NMN in the column is very complex probably because of the bipartite nature of its charges so that subtle differences in extraction and column conditions significantly affect the reliable and accurate detection of NMN. 3. NMN Supplementation: A Shield Against Doxorubicin-Induced Multi-Organ Fibrosis

Introduction In clinical settings, doxorubicin (DOX) is an effective broad-spectrum anti-cancer drug in chemotherapy for various tumors, including hematological malignancies and solid tumors such as leukemia, lymphoma, breast, ovarian and lung cancer. However, there is a formidable obstacle for the wide application of DOX, namely its toxic side effects on vital organs such as heart, liver and lung. Intriguingly, the toxic side effects of DOX are intricately linked to the involvement of nicotinamide adenine dinucleotide (NAD+). To this end, supplementing nicotinamide mononucleotide (NMN), an NAD+ precursor, may be an effective approach to mitigating the toxicity of DOX. About DOX DOX, which is isolated from a soil bacterium, Streptomyces peucetius, was discovered in 1969 as a homolog of daunorubicin. Despite of its potent efficacy, long-term administration of DOX can cause severe damage to numerous organs in the body, with the heart being the primary target of doxorubicin toxicity. DOX has dose-dependent cardiotoxicity, including left ventricular dysfunction, progressive left ventricular remodeling and heart failure. Importantly, the accumulation of DOX can cause fibrosis which plays key roles in pathological development of multiple organs. Multi-organ injury induced by DOX in mice DOX leads to the toxicity to multiple organs, with the most prominent impacts upon heart, liver and lung. Morphologically, decreased thickness in the left ventricular posterior wall (LVPW), destroyed myocardium architecture, edema in hepatic cells and collapsed alveolar spaces are observed following DOX treatment. Besides, the reduced body weight (~20%) and organ weights, along with high mortality rate (60%), are viewed in post DOX injection. Moreover, NMN results in the reduced absolute number of hematopoietic stem/progenitor cells (HSPCs), short-term hematopoietic stem cells (ST-HSCs) and multipotent progenitors (MPPs).
